ZnT8 autoantibodies are important serological markers for diagnosing type 1 diabetes mellitus (T1D). ZnT8 is found in pancreatic beta cells and plays a role in regulating insulin secretion. ZnT8 autoantibodies primarily target the C-terminal domain of ZnT8 (residues 268-369). In
the human population, gene polymorphism at the codon for the 325th amino acid leads to the expression of three protein variants: Arginine (R) 325, Tryptophan (W) 325, and rarely Glutamine (Q) 325. ZnT8 Ab can be specific to the R325 or W325 variant or non-specific to residue 325. Sera that reacts exclusively with the Q allele is extremely uncommon  The IMD ZnT8 Ab ELISA is capable of detecting and quantifying autoantibodies specific to R325, W325, or non-specific to residue 325 variants.
