  • 介紹

    胰島素是一種僅在胰腺 β 細胞中產生的肽激素。它由A鍊和B鏈組成,通過兩個二硫鍵連接。
    胰島素是負責葡萄糖代謝的主要激素。胰島素分泌受損和胰島素抵抗是 2 型糖尿病 (T2D) 的關鍵原因。

  • 測定原理

    該測定是雙位點ELISA。微孔板預先塗有抗胰島素的單克隆抗體。將標準品和样品加入孔中,並與與辣根過氧化物酶 (HRP) 酶偶聯的單克隆抗體共同孵育。在去除任何未結合物質的洗滌步驟之後,加入 TMB 底物,顏色與最初結合的胰島素量成比例。停止測定並使用微孔板讀數器測定孔的光密度。由於吸光度的增加與捕獲的胰島素量成正比,因此可以從每個測定中包含的參考曲線中插入未知樣品濃度。

    B. 敏感性

    該測定法可測量的最低胰島素水平為 3 µU/mL。

    D. 恢復
